If you are wanting to back up all the folders in the site without wp-content, then just tick the box for "WordPress core (including any additions to your WordPress root directory)". You don't need to tick "Any other directory on your server that you wish to back up" or to enter a directory there, because that option is to *also* backup further folders from elsewhere. But what you want is covered by "WordPress core (including...".
